A comparison of the effect of omeprazole and rabeprazole on clozapine serum concentrations.
Proton pump inhibitors (PPIs) such as omeprazole are used commonly in the treatment of gastrointestinal disorders caused by clozapine, the novel atypical antipsychotic in the treatment of refractory schizophrenia. The effective dosage of clozapine varies widely due to variation in metabolism, and more specifically CYP1A2 activity. The effect of rabeprazole, a new PPI, on CYP1A2 activity and clozapine concentrations is not known. This project aims to determine whether rabeprazole has an affect similar to omeprazole on clozapine concentrations. Also, we investigate the pharmacokinetic responses of rabeprazole on CYP1A2.
