Chemokines, mast cell and airways smooth muscle in asthma

In this current research, I'm trying to find out why there is a difference in the production of certain chemokines from the airway smooth muscle of asthmatics, particularly IP-10 and ITAC. we are hypothesizing that there is an alteration in the signalling pattern between asthmatic and non-asthmatics, therefore, we're using selective inhibitory agents that would act on major molecules in the signaling cascade and ultimately inhibit chemokines production. Also, we're extracting mRNA and performing supernatant collection and protein assays in an attempt to understand and logically explain the difference on molecular and DNA level between the two cases.
